Abstract

This chapter summarizes major methods and applications of protein synthesis in biochemistry. The use of protein thioesters generated with inteins for use in expressed protein ligation, cysteine modification strategies, and enzyme catalyzed approaches have been highlighted. More recent applications of these methods to analyze the influence of posttranslational modifications on protein structure and function have been emphasized.
